اجزای شیمیایی، سمیت تدخینی و تاثیر بیوشیمیایی و ضد تغذیه‌ای اسانس Eucalyptus oleosa Muell. روی سوسک کشیش، Rhyzopertha dominica (F.)

چکیده

سوسک کشیش، Rhyzopertha dominica (F.)، باعث ایجاد خسارات کمی و کیفی گسترده­ای به دانه­های غلات انباری می­شود. استفاده از ترکیبات شیمیایی ساختگی در مدیریت چنین آفاتی آثار جانبی زیادی از قبیل آلودگی محیط زیست، تهدید سلامتی انسان و توسعه مقاومت آفات را در پی داشته است. توانمندی حشره­کشی نویدبخش اسانس­های استخراج شده از گونه­های مختلف جنس Eucalyptus روی حشرات آفت در پژوهش­های اخیر نشان داده شده است. در این تحقیق، تاثیر کشندگی و زیرکشندگی اسانس E. oleosa Muell. روی حشرات کامل R. dominica بررسی شد. تجزیه شیمیایی اسانس نشان داد که ترکیبات ترپنی نظیر 1,8-cineole، α-pinene، trans-pinocarveol، (S)-α-terpineol و β-eudesmol واجد بالاترین مقدار بودند. حشرات کامل در برابر تدخین اسانس حساس بودند؛ به­طوری­که مقادیر LC50 (غلظت کشنده 50 درصد) محاسبه شده بعد از 24، 48 و 72 ساعت به­ترتیب 923/42، 828/35 و 044/34 میکرولیتر بر لیتر هوا برآورد شد. با وجود افزایش فعالیت آنزیم­های آلفا- و بتا-استراز، محتوای پروتئین، گلیکوژن و لیپید کل و فعالیت­های آمیلولیتیک و پروتئولیتیک حشرات کامل تیمار شده با LC30 (غلظت کشنده 30 درصد) اسانس به صورت معنی­داری در مقایسه با شاهد کاهش یافت. شاخص مصرف (CI)، نرخ مصرف نسبی (RCR) و نرخ رشد نسبی (RGR) هم در حشرات کامل تیمار شده به­صورت معنی­داری کاهش یافت. شاخص بازدارندگی تغذیه­ای (FDI) 40/59 درصدی تحت تاثیر غلظت کشنده 30 درصد اسانس در حشرات تیمار شده محاسبه شد. با توجه به تاثیر کشندگی و زیرکشندگی مشاهده شده، اسانس E. oleosa می­تواند برای انجام بررسی­های تکمیلی در راستای مدیریت R. dominica مورد توجه قرار گیرد.

The lesser grain borer, R. dominica, causes extensive quantitative and qualitative loses on stored cereal grains. The use of synthetic chemicals in the management of such insect pests resulted in several side effects such as environmental contamination, potential impacts on human health and development of pest resistance. Promising insecticidal potential of the essential oils isolated from several species of Eucalyptus genous against insect pests was shown in recent study. In this study, the lethal and sublethal effects of E. oleosa essential oil were investigaetd against the adults of R. dominica.  Chemical analysis of the essential oil indicated that terpenes, including 1,8-cineole, α-pinene, trans-pinocarveol, (s)-α-terpineol, and β-eudesmol, had high amount. The adult insects were susceptible to the fumigation of essential oil so that LC50 (Lethal Concentration to kill 50% of insects) values were estimated as ‎42.923, ‎35.828, and ‎34.044 µl/l air after 24, 48 and 72 h, respectively. Despite increases in a- and β-estearse enzymes, total protein, glycogen, and lipid contents and digestive amylolytic and proteolytic activities of the adults treated with LC30 (Lethal Concentration to kill 30% of insects) of essential oil were significantly decreased in comparision with the control. The consumption index (CI), relative consumption rate (RCR), and relative growth rate (RGR) were also significantly decreased in treated adults. A 59.40% feeding deterrence index (FDI) was evaluated for insects treated with LC30 of  the essential oil. According to observed lethal and sublethal effects, the essential oil of E. oleosa can be considered in further investigations in the management of R. dominica.
