IN VITRO PROPAGATION OF CISTUS CRETICUS L.

Abstract:
Cistus creticus L. (Cistaceae), a species with medicinal and aromatic properties, was propagated in vitro. After a heat treatment in an autoclave at 100°C for 20 min to overcome hardseededness, seeds were disinfested with calcium hypochlorite (1.5% for 20 min) and ethanol (70% for 1 min) and placed on the modified WPM. Shoot formation was obtained 30 days after the first subculture, from which new shoot segments were taken. The segments, subcultured on the same medium, were rooted alone or after supplementing the medium with different concentrations of IBA (0.98, 1.97 and 3.94 µM) or NAA (0.1 and 0.5 µM). Zeatin (0.2 and 0.5 mg l-1) was used for callus induction. The plantlets were successfully acclimatized to ex vitro conditions.
