カンショにおけるショ糖合成酵素の発現様式に関する研究

過去に報告されたショ糖合成酵素(EC 2. 4. 1. 13)で保存性の高い領域をもとにして一本鎖のオリゴヌクレオチドプライマーを作成し, カンショ塊根のポリ(A)+ RNAより合成したcDNAを鋳型としてPCRを行ったところ, 1,191bpのcDNA (IBSUS)が増幅された. IBSUSの塩基配列はバレイショの二種類のショ糖合成酵素cDNAの対応する領域と82%および86%の高い相同性を示した. 単子葉植物であるトウモロコシ, イネおよびオオムギのショ糖合成酵素とは76~77%とわずかに相同性が低かった. カンショ塊根におけるショ糖合成酵素の活性は, 葉身, 葉柄, ほふく茎および細根のショ糖合成酵素活性に比べて高かった. また, 塊根形成過程において, 根のショ糖合成酵素活性はショ糖含量の上昇にともなって著しく増加した. IBSUSをプローブとしたノーザンブロット分析では, 葉柄, ほふく茎, 細根および塊根において約2.4kbの一本のショ糖合成酸素mRNAが検出され, 部位別および塊根形成過程における根のショ糖合成酸素のmRNAレベルはショ糖合成酵素活性で得られた結果とよく一致した. また, 葉柄ではショ糖濃度を高めるとショ糖合成酵素の活性が増加した.
