Krebs-Henseleit (KH) Buffer

118 mM NaCl

4.7 mM KCl

1.2 mM MgSO4

1.25 mM CaCl2

1.2 mM KH2PO4

25 mM NaHCO3

11 mM glucose

Make one liter fresh on the day of isolation from 20x stocks (except the glucose). KH buffer has to be oxygenated by gassing the solution with 95% O2/5% CO2 until the pH is apporximately 7.4.

If difficulty is encountered with the quantity and/or quality of the isolated cells, which can be caused by variations in water quality, Joklik's media (GIBCO/BRL) can be used in place of calcium-free KH buffer.
