A Pepsin Pancreatin Digest Index of Protein Quality Evaluation
Abstract
A pepsin pancreatin digest index was devised for a rapid, accurate estimation of protein quality. The index was calculated from the amino acids released by an in vitro digestion with pepsin followed by pancreatin. The amino acids were determined by automatic amino acid analysis which allowed rapid protein quality evaluations with very small samples. Using whole egg as a standard, excellent correlation was observed between the pepsin pancreatin index values for 12 proteins and their biological values reported in the literature from feeding trials. The proteins tested were selected to cover a wide range of protein quality. The pepsin pancreatin digest index values showed better correlation with the biological values for the growing rat than did the essential amino acid index which tended to overestimate the value, or the chemical score which tended to underestimate it.

Role of food processing and incorporating legumes in food products to increase protein intake and enhance satiety
2024, Trends in Food Science and TechnologyLegumes are an important source of plant-based protein that are known to provide many beneficial health effects, but their global consumption is low. Thus, processing legumes to improve their protein digestibility and incorporating them into commonly consumed foods could be a useful strategy to increase legume consumption and obtain the nutritional benefits, including increased protein intake and enhanced satiety.
This review highlights various food processing techniques that can be utilised to enhance the digestibility of legume protein, particularly when used to enrich food products. Whether such a strategy benefits metabolic health, satiety perception, and postprandial hormone responses to such foods is then evaluated. Finally, consumer acceptance of legume-based food products is examined.
Food processing techniques can be used to enhance the digestibility of legume protein, and this effect is evident when incorporating legume proteins into food products. The increased levels of protein in such foods are found to be more satiating by consumers, while also eliciting improved postprandial hormone responses, suggesting they would aid in improving metabolic health. The incorporation of legumes into foods may reduce the consumer acceptance of such products. However, by utilizing a combination of food processing techniques, the sensory properties of legumes can be effectively manipulated to improve the acceptability of legume-enriched food products. Thus, the incorporation of legume protein into foods appears to be a viable strategy to encourage legume consumption and improve overall health.
Physicochemical, structural, and functional characterization of guar meal protein isolate (Cyamopsis tetragonoloba)
2024, HeliyonGuar korma and churi protein isolates were assessed for their physicochemical, nutritional, functional, structural, and digestibility properties for their application in the food industry. The water extracted protein isolate of guar korma showed a protein content of 89.7 % and a yield of 48.7 %. Water extracted protein isolate of guar korma showed an excellent protein efficiency ratio, essential amino acid/total amino acids (34.35 %), amino acid score, and protein digestibility corrected amino acid score values, suggesting the existence of high-quality proteins. Water extracted protein isolate of guar korma contains all the essential amino acids except Methionine and Cysteine, according to World Health Organization recommendations for children and adults. The protein profiling of water extracted protein isolate of guar korma was analyzed using 12 % sodium dodecyl sulfate polyacrylamide gel electrophoresis and indicated the presence of eight major protein bands in the range of 17–100 kDa. In vitro digestibility of water extracted protein isolate of guar korma showed the complete digestion of the abundant protein bands within 15 min. Further, the foaming capacity, water/oil holding capacity, and emulsifying stability of water extracted protein isolate of guar korma were comparable with soy protein isolate. Fourier Transform Infrared and Circular Dichroism spectral analysis revealed the presence of several aromatic groups and β-sheets, random coils respectively in water extracted protein isolate of guar korma. The morphological nature of the guar protein isolate was characterized by Scanning Electron Microscopy. Overall, these findings support that water extracted protein isolate of guar korma has excellent functional and nutritional properties and could be a potential alternative plant protein in food industries.
Effect of soaking and germination on grain matrix and glycaemic potential: A comparative study on white quinoa, proso and foxtail millet flours
2023, Food BioscienceThis study investigated the effect of soaking and germination treatment on interactions among matrix components and functional characteristics of white quinoa, proso and foxtail millet flour. The millet and white quinoa grains were soaked, germinated and dehulled for analysis. Soaking and germination significantly (p < 0.01) affected the content of total starch, amylose, amylopectin, total polyphenols and total flavonoids. A significant variation (p < 0.01) in in-vitro protein digestibility and glycaemic potential of proso, foxtail millet and white quinoa flour was observed after soaking and germination. SEM images and ATR-FTIR spectra revealed alteration in morphology and amorphous and crystalline regions of germinated flours. Principal component analysis revealed less similarity among the germinated millet and white quinoa flour in terms of matrix components and functional characteristics. Hence germinated flours of white quinoa, proso and foxtail millet could be utilized as a beneficial constituent in food products.
The local barley cultivar Nure was subjected to a controlled sprouting process, obtaining flour and grains that were fermented to produce type II sourdoughs. The germination process led to the reduction of starch and total dietary fibers (−58 and −48% compared to plain barley flour) and to the increase of phenolic compounds and protein bio-accessibility. Fermentation of sprouted barley (SB) with selected lactic acid bacteria determined a further enhancement of its nutritional features, by means of the increased free amino acids (up to 35%) and γ-aminobutyric acid concentrations (up to 57%), and decreased phytic acid content. The potential of SB sourdough to be used as ingredient in bread making was then investigated. Overall, the negative effects (on dough rheology and baking performance) related to the intense enzymatic activities characterizing the sprouted barley flour are strongly mitigated by the fermentation process. Therefore, besides improving bread nutritional and technological attributes, the use of SB sourdoughs, by supplying the native enzymes present in the sprouted grains, but in a less invasive form, could help decrease or substitute the use of commercial enzymes or flour improvers commonly used in the baking industry.
To improve the utility value of faba bean meal (FBM), the effects of solid-state fermentation (SSF) by a strain of Bacillus pumilus on nutritional value, in vitro protein digestibility, anti-nutritional factors (ANFs), flavonoid, phenols and antioxidant activity were evaluated. The results revealed that the concentrations of ANFs (phytic acid and proanthocyanidin) were significantly decreased (P < 0.05) after fermentation. Fermented FBM showed higher nutritional value. Crude protein, amino acids and trichloroacetic acid-soluble protein contents in fermented FBM were higher than those from unfermented FBM. Meanwhile, the contents of phenols and flavonoid were remarkably (P < 0.05) increased after SSF. Scanning electron microscope showed that the surface of fermented FBM was more irregular and rougher than unfermented FBM. Moreover, SSF treatment significantly (P < 0.05) increased DPPH and superoxide anion radical scavenging capacities. The results indicated that SSF of FBM with B. pumilus is a promising and efficient method to improve nutritional value and antioxidant activity, as well as decrease the concentration of ANFs, which also endow the potential value of FBM on replacing soy bean as primary protein source in the feed ration of different animals.
Effect of Piaractus brachypomus fish protein hydrolysate on physicochemical, sensory, and storage properties of cookies
2023, Biocatalysis and Agricultural BiotechnologyThe impact of different concentrations of red-bellied pacu fish meat protein hydrolysate (RPMPH) on the physicochemical, sensory, and storage properties of cookies was studied. RPMPH was prepared from red-bellied pacu fish meat by probiotic Bacillus fermentation for 48 h. RPMPH showed 87.48% protein content with DPPH scavenging activity (71.22%), ABTS scavenging activity (90.05%), ferric reducing antioxidant power (278.13 μM TE/g), and Fe2+ chelating activity (65.46%). Cookies incorporated with RPMPH displayed a higher protein content (11.76–14.40%) and in vitro protein digestibility (65.21–96.0%) compared to control (without RPMPH) cookies (P < 0.05). The total dietary fiber content of all cookies ranged from 1.74 to 2.63 g/100 g sample. The control-cookies had a higher diameter (7.0 mm) and spread ratio (8.37%) compared to cookies incorporated with RPMPH (P < 0.05). The cookies incorporated with RPMPH exhibited a slightly higher water absorption index (6.79–7.51%), water solubility index (2.74–2.88%), and hardness (41.36–49.48 N) than the control-cookies. Color of cookies, influenced by incorporation of RPMPH. Further, the cookies incorporated with RPMPH showed slightly higher moisture and water activity compared to control-cookies throughout the storage (P < 0.05). Conversely, cookies incorporated with RPMPH displayed lower free fatty acids, peroxide values, and thiobarbituric acid substances than control-cookies (P < 0.05) throughout the storage period. The cookies with 8% RPMPH exhibited a higher score for overall acceptability and presented a better amount of essential amino acids. The present study suggested that 8% RPMPH can be incorporated into the cookie formulation, enhancing the nutritional profile and storage stability.
