This Letter to the Editor summarizes our response to comments by Tseng and Yang (2017) concerning our recent publication in the Journal of Medical Entomology (Seri Masran and Ab Majid 2017).
Tseng and Yang (2017) discussed four aspects of our paper (Seri Masran and Ab Majid 2017), and we have organized our response to address each of these in parallel.
1. Sequence data. We pooled three individuals of the tropical bed bug from the same site into a single DNA mixture and then subjected this mixture to PCR and sequencing. The rationale for sequencing these small pools rather than individuals was to ensure greater DNA quantity and quality and better sequencing output and to reduce cost by reducing the number of sequencing reactions. Using DNA pooling rather than individuals has been done previously on honeybees by Haberl (1999), termites by Cheng et al. (2013), pea aphid by Zhang and Hewitt (1997), grasshoppers by Zhang et al. (1995), weevils by Gillett et al (2014), stored grain pests by Schlipalius et al. (2001), beetle larvae by DiZinno et al. (2002), potato psyllids by Nachappa et al. (2011), and many more.
