Discovery of an androgen receptor modulator pharmacophore based on 2-quinolinones
Graphical abstract
A series of alkylamino-2-quinolinone compounds (3) was discovered as androgen receptor modulators based on an early linear tricyclic quinoline pharmacophore (1). The series demonstrated selective high binding affinity to androgen receptor and potent receptor modulating activities in the cotransfection assays.

However, relatively few 1H-quinolin-2-one derivatives have been reported as antibacterials [17]; among them, the 4-substituted analogs rationally designed by the group of Jayashree, based on docking experiments with bacterial DNA gyrase [18].

Several carbostyril derivatives are also nitric oxide production inhibitors (Ito et al., 2004), 5-HT1B/5-HT2A receptor antagonists (McCort et al., 2001), Rho-kinase inhibitors (Letellier et al., 2008), androgen receptors (Oeveren et al., 2007), p38αMAP kinase inhibitors (Peifer et al., 2008) and induce mitochondrial dysfunction (Chilin et al., 2005). Therefore, the synthesis of 2-quinolone derivatives has kindled a strong interest in us.
