Nε-Thioacetyl-lysine: A multi-facet functional probe for enzymatic protein lysine Nε-deacetylation

Peptides containing Nε-thioacetyl-lysine and Nε-acetyl-lysine were evaluated for their de(thio)acetylation catalyzed by human HDAC8 and SIRT1, two distinct protein deacetylases. Nε-Thioacetyl-lysine was found to be a mimic of Nε-acetyl-lysine for HDAC8-catalyzed reaction, but to confer inhibition against SIRT1.

Profiling sirtuin activity using Copper-free click chemistry

They are thioacyllysine peptide-based probes [36] with either a benzophenone or a diazirine photoaffinity group, and a terminal alkyne “clickable” tag. They demonstrated good labeling efficiency and isoform selectivity in labeling individual recombinant sirtuins, and one or more isotypes in protein mixtures or cell lysates.
Development of activity-based probes for the protein deacylase Sirt1
2020, Bioorganic ChemistryCitation Excerpt :Samples were incubated for 10 min at ambient temperature and then subjected to UV-irradiation and CuAAC followed by in-gel fluorescence analysis as described in Section 2.3. Using a modular peptide-based approach, two potential sirtuin activity-based probes (ABPs) were synthesized, each consisting of four elements: (1) Sirt1 substrate peptide sequences based on either histone 3 (H3K14) [37] or p53 [13,14,17]; (2) a thioacetyl-lysine residue for mechanism-based sirtuin inhibition [25,26]; (3) a diazirine photolabeling group for efficient crosslinking to Sirt1 when bound [38,39]; and (4) an alkyne reporter tag amenable to copper(I)-catalyzed alkyne-azide cycloaddition (CuAAC) labeling [40] with either a fluorophore or biotin for detection and measurement of Sirt1 activity (Fig. 1B-D). The development of the sirtuin ABP workflow into an efficient and reproducible methodology required multiple rounds of optimization.
