Two novel side chains which had previously been found to enhance antagonist activity in the dihydrobenzoxathiin SERM series were applied to three existing platforms. The novel side chains did not improve the antagonist activity of the existing platforms.

Lewis acid promoted coupling [32] of 31b and 31c with methyl-1,2,3,4-tetra-O-acetyl-d-glucopyranuronate provided the β-anomers 37 and 38 in acceptable yields. Hydrolysis of the β-anomers using LiOH, and following treatment with tetrabutylammonium fluoride (TBAF) furnished the desired 4′- and 6-β-glucuronides 39 and 40 in acceptable yields.
Pharmacophore and QSAR modeling of estrogen receptor β ligands and subsequent validation and in silico search for new hits

The literature was extensively surveyed to collect as many structurally diverse ERβ ligands as possible (1–119, see Table A and Figure A in Supporting Information) [2,3,6,8,9,11,16,17,39–55]. The collected compounds were carefully selected from around 800 reported inhibitors in such a way to guarantee dissimilar affinities to ERα and ERβ, and therefore, to allow access to selective ERβ pharmacophore models.
