Abstract

Targeting the IL17 pathway and more specifically the nuclear receptor RORγ is thought to be beneficial in multiple skin disorders. The Letter describes the discovery of phenoxyindazoles and thiophenoxy indazoles as potent RORγ inverse agonists. Optimization of the potency and efforts to mitigate the phototoxic liability of the series are presented. Finally, crystallization of the lead compound revealed that the series bound to an allosteric site of the nuclear receptor. Such compounds could be useful as tool compounds for understanding the impact of topical treatment on skin disease models.

    • Modes of action insights from the crystallographic structures of retinoic acid receptor-related orphan receptor-γt (RORγt)

2023, European Journal of Medicinal Chemistry
      Citation Excerpt :

These MD insights have reasonably explained the observed different conformations of W317 in the reported x-ray structures as discussed above, and demonstrated their inverse agonism through the tuning the side chain of W317 in RORγt. While great efforts were spent on the design of inverse agonists binding at the orthosteric binding pocket at RORγt LBD, a new type of inverse agonists were discovered to bind at the allosteric site (Figs. 2B and 12) [55–60,87], which occupies the site originally for the coactivator in the apo-state and active RORγt receptor (Figs. 2A and 3A). This site is formed by the helices H4, H5, H11 and H12.

French arms of Galderma R&D and NovAliX advanced the development of indazole compounds, as disclosed in a 2016 patent by Merck. They optimized these compounds for stability, making them suitable for topical formulation for the treatment of psoriasis, and evolved a series of phenoxyindazoles (5LWP) and phenylthioindazoles.37 AstraZeneca has made a huge contribution to knowledge in the area of RoR-γt crystallography.

    • Delineation of the molecular determinants of the unique allosteric binding site of the orphan nuclear receptor RORgt

      2020, Journal of Biological Chemistry
      Citation Excerpt :

Whereas other amino acids that delineate the allosteric pocket can be replaced for those with other side chains, modifications to these two elements are detrimental. Thus, the results of this study strongly bring forward that the allosteric pocket, as present in RORγt and receptive to MRL-871 and other ligands (16, 27–30, 37), is highly unique to RORγt within the NR superfamily. This implies that RORγt is the only NR capable of forming this specific type of allosteric pocket, which is also supported by a commercially available panel of cell-based NR reporter assays against which an MRL-871 analog was tested (16).
