Elucidating the potential neurotoxicity of chiral phenthoate: Molecular insight from experimental and computational studies
2020, ChemosphereCitation Excerpt :These points have been verified firmly by the experimental results of Li et al. (2002, 2003, 2007, 2010). In previously toxicological risk studies, some researchers have discovered that the racemic phenthoate can trigger a number of noxious effects in mammals (mouse and rat), e.g. allergic disease, cumulative toxicity, lung damage, mutagenicity, oxidative stress, and prenecrotic changes in the tissue (Saigal et al., 1982; Gupta, 1984; Nahas et al., 1997; Sato et al., 1998; El-Shenawy and Al-Ghamdi, 2014), but the neurotoxic effects of phenthoate enantiomers on the body remains ambiguous, and more importantly, acetylcholinesterase (AChE) as the biological target for organophosphate in living organism, the enantioselective role of such chiral factor (AChE) in the neurotoxicity of phenthoate is yet an unsolved mystery. Given the above-mentioned research background, this attempt was to deeply explore the molecular/atomic processes of the toxic reaction of nerve AChE enzyme with chiral phenthoate at the enantiomeric level, and then elaborate the molecular basis for neurotoxicological effects of phenthoate enantiomers on the human body.
