Identification of a particular antigen from a parasite cDNA library using antibodies affinity purified from selected portions of Western blots

Abstract

Portions of nitrocellulose filters containing blotted electrophoresed antigens of Schistosoma japonicum adult worms were reacted with polyclonal rabbit antisera raised to this human parasite. Eluted antibodies were used as probes for detection of antigen-positive clones in an Escherichia coli λgt11 amp3 expression library of adult worm cDNA. Several cloned antigens corresponding to a S. japonicum antigen of Mr 26 000, being sought as a candidate vaccine molecule in a mouse model of schistosomiasis japonica, were identified using this approach. The method provides an antibody reagent that is an attractive alternative to other more tedious means of producing oligospecific antibodies, including monoclonal antibodies, for screening of expression libraries.

    Immunomics involves using cloned segments of genomic DNA for genetic immunization to obtain a large collection of antibodies and to generate microarrays of these antibodies for tracing differentially expressed cellular proteins (Klysik, 2001). With the aim of identifying the parasite proteins that interact with the host's immune system, various serum samples from infected and vaccinated animals were employed to identify parasite immunogenic proteins (Beall and Mitchell, 1986; Goudot-Crozel et al., 1989; Jeffs et al., 1991; Lanar et al., 1985; McManus et al., 1998). The combination of proteomics and serology has some advantages over traditional expression library screening, partly because the latter cannot detect immunogenic epitopes arising from post-translation modifications.
