Abstract

A method for identifying monocytes by the ‘non-specific esterase’ stain is described. This method is particularly applicable to mononuclear cell suspensions obtained by Ficoll-Hypaque density gradient separations and allows rapid as well as accurate determinations.

Evaluation of the effect of interleukin-10 on the multiplication of Mycobacterium avium complex in human macrophages and in C57BL/6 mice

To examine the effect of recombinant human IL-10 (rhlL-10) on MAC infection of human macrophages and C57BL/6 mice.
We compared rhlL-10 with the effects of the immunosuppressive drugs prednisolone and cyclosporin A, both in vitro and in vivo.
There was no effect of rhlL-10 on the multiplication of MAC in human macrophages after 1 week of infection. In C57BL/6 mice, rhlL-10 at 2.5 or 25 μg/mouse had no additional multiplicatory effect after 3 weeks of infection, while the spleens of mice treated with prednisolone had 600% higher bacteria than controls or rhlL-10-treated mice (p<0.01).
These data suggest that rhlL-10 does not further decrease the resistance of human macrophages and C57BL/6 mice to MAC infection, whereas prednisolone leads to increased multiplication of MAC in the spleens of infected C57BL/6 mice. These results may be of interest in the context of the therapeutic use of rhlL-10 in some autoimmune disorders.
Transferrin and iron salts modulate differently tumor necrosis factor- α secretion by cultured human mononuclear cells

Iron is an essential element for the normal function of many biological processes, including the immune response and hematopoiesis. Since iron deficiency results in impaired cytokine production (TNF-α), we investigated if iron, bound to its transport protein transferrin or by itself, modulates the secretion of tumor necrosis factor-α (TNF-α) by circulating monocytes. In particular, we examined the effect of Tf-bound iron or ferric chloride (FeCl₃) in the secretion of TNF-α by cultured blood mononuclear cells (BMNC) obtained from women with either normal Fe status (n=10) or with iron deficiency (n=10). The addition of 30 μM Tf, either in its apo or holo form, to BMNC cultures derived from both normal and iron deficient subjects, induced a marked increase in TNF-α secretion by cells, to about 1.2–1.4 ng/mL. Similarly, the addition of μM amounts of FeCl₃ to normal BMNC resulted in a dose-dependent increase of TNF-α secretion. By contrast, BMNC from iron deficient subjects were unable to secrete TNF-α under similar conditions. Lipopolysaccharide (LPS) induced a maximal secretion of TNF-α (3.9 ± 0.6 ng/mL, mean ± SEM) in BMNC derived from both normal and iron deficient women, an indication that ID cells had the capacity to secrete TNF-α in response to a bacteriologic insult. However, the combined addition of LPS and iron-salt did not induce a further increase in TNF-α secretion. These findings indicate that iron modulates the in vitro secretion of TNF-α by human mononuclear cells through a process that depends both on the iron status of the subject and on the form in which iron is supplied. Moreover, transferrin also induces the secretion of TNF-α in a way apparently independent of its iron-donating capacity.
Absence of involvement of nitric oxide in LP-BM5-induced immunodeficiency syndrome

To examine the role of nitric oxide (NO) in murine AIDS (MAIDS) pathogenesis, we determined NO production and inducible NOS (iNOS) mRNA expression in the macrophages of LP-BM5-infected mice, together with the in vivo effects of l-NAME, a competitive inhibitor of NO synthase. LP-BM5 infection induced neither spontaneous nitrite production nor iNOS mRNA expression. No differences in IFNγ + LPS-induced nitrite production or iNOS mRNA expression were observed in macrophages from non-infected or infected mice. Spleen weight, ecotropic MuLV replication, the blood lymphocyte phenotype and proliferative response of splenocytes were not modified by l-NAME. LP-BM5 infection did not increase macrophage NO production and NO production did not appear to protect against LP-BM5-induced immunodeficiency.
Tissue factor expression in human leukemic cells

Patients with acute leukemia are at increased risk for thrombotic and hemorrhagic complications, particularly those patients with acute promyelocytic leukemia (APL) undergoing induction chemotherapy. These serious complications have been attributed by some authors to the release of tissue factor (TF) procoagulant activity (PCA), particularly during cytotoxic chemotherapy.
In previous studies of normal peripheral blood cells, only cells of the monocyte lineage have been found to express TF PCA. Therefore, several questions remain regarding the origin and characterization of the PCA in malignant leukemic cells, particularly those thought to be derived from granulocyte progenitor cells. We utilized a full-length cDNA probe, several monoclonal antibodies (MAbs) and a sensitive one-stage PCA assay to study the expression of TF in the human leukemia cell line, HL-60, in human peripheral blood monocytes/macrophages ( $Mo Mø$ ) and in highly purified populations of human polymorphonuclear leukocytes (PMN).
In the HL-60 cells we detected low but significant levels of TF mRNA and TF antigen (TF:Ag). In unstimulated cells, coordinate increased levels of TF mRNA, TF:Ag and TF PCA expression were noted following phorbol-ester-induced macrophage differentiation of the cells, but a decreased level of TF mRNA with no change in the basal level of TF:Ag expression occurred following retinoic acid-induced granulocyte differentiation of this cell line. Long-term cultures of stimulated mature $Mo Mø$ demonstrated initial coordinate expression of TF mRNA, TF:Ag and TF PCA, but TF:Ag expression persisted even after 7 days (when TF PCA was undetectable). No TF PCA, TF:Ag or TF mRNA was demonstrated in highly purified populations of human PMN, regardless of culture conditions.
Discordant expression of TF mRNA, TF:Ag and TF PCA in HL-60 cells suggests the possibility of novel, post-synthetic mechanisms for the regulation of TF PCA expression, which might be dependent on the phenotypic differentiation level of the cell. Such mechanisms (yet to be defined) might account for the ability of some leukemic cells, which frequently express characteristics of more than one cell line (e.g. monocytes and granulocytes), to express a TF gene product capable of activating blood coagulation.
