Untersuchungen über das schicksal , die verteilung im organismus des peroral oder parenteral verabreichten vitamin B12

Zusammenfassung

  • 1.

    1. In Fortsetzung von Untersuchungen mit radioaktivem Vitamin B12 (60Co) wurde festgestellt, daß auch bei langfristiger Versuchsanordnung über 14 Tage lang mit täglichen Gaben physiologischer Vitamin B12-Mengen (0.05 Gamma) subcutan oder peroral, im Nierengewebe eine auffallend starke Anhäufung dieses Vitamins stattfindet.

    2. Die erhobenen Befunde zeigen, daß die protrahierte Zufuhr geringer Mengen Vitamin B12 die physiologische ist und den einmalig verabreichten Riesendosen (1000 Gamma Vitamin B12) vorzuziehen ist.

    3. Daraus wird geschlossen, daß die Niere das in den Primärharn ausgeschiedene Vitamin B12 rückzuresorbieren vermag.

  • 4.

    4. Die Vitamin B12-Aufnahme der Leber hat sich im Vergleich mit dem Vitamin B12-Retentionsvermögen der Nieren als viel geringer erwiesen.

  • 5.

    5. Auch das Nervengewebe (Gehirn) und das Myocard wiesen ein starkes Aufnahmevermögen von Vitamin B12 auf, wogegen dieses Phänomen sich autoradiographisch in der Skelettmuskulatur höchstens andeutungsweise nachweisen ließ.

  • 6.

    6. Diese experimentellen Befunde erhellen die bedeutende Aufgabe, welche dieses Vitamin im Stoffwechsel des Nervengewebes und des Myocards zu erfüllen hat und stehen im Einklang mit den klinischen Beobachtungen bei Vitamin B12-Mangelzuständen (funiculäre Myelose, sowie Myocardose, nach Wuharmann11).

Abstract

  • 1.

    1. In the course of continued research with radioactive vitamin B12 (60Co) it was established that with long-term experiments lasting over 14 days and involving administration of daily physiological doses of vitamin B12 (0.05 γ) subcutaneously or orally, a strikingly large quantity of this vitamin accumulates in the kidney tissue.

  • 2.

    2. The results obtained show that the protracted supply of smaller quantities of vitamin B12 is physiologically correct and is to be preferred to single huge doses.

    3. It can thus be concluded that the kidney is capable of reabsorbing the vitamin B12 excreted in the primary urine.

    4. The vitamin B12 absorption of the liver is shown to be much less in comparison with the vitamin B12 retention capacity of the kidney.

    5. Both the nerve tissue (brain) and the myocardium show a great ability to take up vitamin B12, whereas this phenomenon could at most only be detected slightly in autoradiography of the skeletal muscle.

    6. These experimental findings throw light on the important task which this vitamin fulfils in the metabolism of nerve tissues and myocardium, and we are in agreement with the clinical observations on conditions of vitamin B12 deficiency (funicular myelosis, as well as myocardosis, according to Wuhrmann11).

    1. En poursuivant nos recherches à l'aide de vitamine B12 radioactive (60Co) nous avons pu démontrer par des expériences de longue durée (14 jours) sur animaux, en appliquant de petites doses de 0.05 μμg par voie parentérale ou perorale, que les tissus rénaux retiennent fortement la vitamine B12.

    2. Nos résultats — en ce qui concerne la posologie — plaident en faveur d'une thérapeutique par petites doses répétées qui sont plus efficaces que l'administration d'une seule forte dose de 1000 μμg ou plus.

  • 3.

    3. Les reins sont donc capables de réabsorber la vitamine B12 excrétée avec l'urine primaire.

  • 4.

    4. La rétention de la vitamine B12 des reins est sensiblement plus élevée que celle du foie.

  • 5.

    5. Le système nerveux central (cerveau) ainsi que le myocarde se caractérisent également par une forte rétention de la vitamine B12; dans les muscles striés, par contre, on ne décèle qu'une radioactivité minime.

  • 6.

    6. Nos résultats expérimentaux font ressortir l'importance du rôle que joue la vitamine B12 dans le métabolisme du système nerveux et du myocarde; ils confirment ainsi les données cliniques (myelose funiculaire, myocardose) dans les carences de vitamine B12.
