Antibacterial activity of the essential oils from the leaves of Eucalyptus globulus against Escherichia coli and Staphylococcus aureus

Abstract

Objective

To examine the in vitro antimicrobial activities of essential oil of the leaves of Eucalyptus globulus (E. globulus).

Methods

The essential oils of this plant were obtained by the hydrodistillation method. The inhibitory effects of this essential oil were tested against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) by using agar disc diffusion and dilution broth methods.

Results

The results obtained showed that essential oil of the leaves of E. globulus has antimicrobial activity against gram negative bacteria (E. coli) as well as gram positive bacteria (S. aureus).

Conclusions

The encouraging results indicate the essential oil of E. globulus leaves might be exploited as natural antibiotic for the treatment of several infectious diseases caused by these two germs, and could be useful in understanding the relations between traditional cures and current medicines.

Eucalyptus globulus leaf-isolated isorhapontin serves as a natural insecticide via acetylcholinesterase inhibition

Acetylcholinesterase (AChE) inhibitors cause insect death by preventing the hydrolysis of the neurotransmitter acetylcholine, which overstimulates the nervous system. In this study, isorhapontin, isolated from E. globulus leaves, was evaluated as a natural insecticide with AChE inhibition at 12.5 μM. Using kinetic analyses, we found that isorhapontin acted as a competitive inhibitor that binds to the active site of AChE. The inhibition constant (K_i) was 6.1 μM. Furthermore, isorhapontin and resveratrol, which have basic skeletons, were predicted to bind to the active site of AChE via molecular docking. A comparison of the hydrogen bonding between the two stilbenes revealed characteristic differences in their interactions with amino acids. In isorhapontin, Trp83, Gly149, Tyr162, Tyr324, and Tyr370 interacted with the sugar moiety. These results suggest that with further development, isorhapontin can be used as an insecticide alternative.
The antimicrobial efficacy of native Australian essential oils in liquid and vapour phase against foodborne pathogens and spoilage microorganisms

Foodborne pathogens and spoilage microorganisms are a source of concern for public health and the food industry. Considering the problem of antimicrobial resistance in microbes, controlling these microorganisms using synthetic antimicrobials is challenging, thus, essential oils are a potential natural alternative. Previous studies have predominantly focussed on the antimicrobial activity of essential oils in the liquid phase. However, in this work we report on the in vitro antimicrobial activity of the liquid and vapour phases of essential oils from native Australian plants Tasmanian mountain pepper (Tasmannia lanceolata), lemon myrtle (Backhousia citriodora), as well as the common herb thyme (Thymus vulgaris) against 11 common food spoilage and pathogenic bacteria and fungi. Initial screening was conducted using the disc diffusion assay. The minimum inhibitory concentration (MIC) of the oils was determined using the broth microdilution assay. The vapour phase antimicrobial activity of the oils was assessed using an inverted petri plate assay and the vapour phase MIC (V-MIC) was quantified using a vapour diffusion assay. GC-MS was adopted to elucidate the compositional profile of the oil in liquid phase. Two extraction methods, namely solid-phase microextraction (HS-SPME) and gas tight syringe headspace sampling (HS) coupled with GC-MS were used for profiling the composition of the volatiles. The findings of this study confirmed the antimicrobial activity of the oils in both liquid and vapour phases, with lemon myrtle and thyme generally showing stronger antimicrobial activity than Tasmanian mountain pepper. The activity in liquid phase was generally higher than the volatiles. However, lemon myrtle and Tasmanian mountain pepper berry volatiles were more effective against Aspergillus niger and Saccharomyces cerevisiae, respectively, relative to the liquid phase highlighting their potential as vapour phase antifungals. In the liquid and gas phases, the major compounds in lemon myrtle were Z-citral (neral) and E-citral (geranial). Polygodial and thymol were the major compounds in Tasmanian mountain pepper and thyme in the liquid phase, respectively. However, the main compounds in their volatiles were p-cymene and γ-terpinene. In conclusion, our research indicates the essential oils' potent vapour phase activity, highlighting their potential application in gaseous diffusion-dependent food applications such as active antimicrobial food packaging.
Green label marinades: A solution to salmonella and campylobacter in chicken products?

The presence of meat-borne pathogens entering the home remains a concern for consumers, despite advances made in improving antimicrobial interventions and systems within the processing line. Naturally antibacterial food ingredients including citrus juice and essential oils have been proven to inhibit the proliferation of microbial growth with varying success.
This study aims to investigate the antimicrobial and sensory effects of mixtures of essential oils, fruit juices and herbs at established Minimum Inhibitory Concentrations (MICs) for their biopreservative effect on general microbiota of chicken and against chicken challenged with selected pathogenic/surrogate microorganisms.
Three marinade compositions were designed for use on chicken meat; lemon juice, thyme oil and black pepper (M1), lime juice, lemongrass oil and chilli paste (M2), and olive oil, oregano oil, basil oil and garlic paste (M3). These marinades were assessed for antibacterial effects against Salmonella enterica, Campylobacter jejuni and Listeria innocua on marinaded chicken drumsticks stored in aerobic conditions at 4 °C. Consumer tasting sessions were also conducted with a small focus group using selected final marinades.
M1 and M2 were effective at significantly reducing initial pathogen carriage from 6 Log CFU/g to 2 Log CFU/g on refrigerated chicken meat as well as increasing the shelf-life of the product during cold-storage from 2 days to 7 days. However, consumer studies indicate that the flavours these marinades impart to treated products can be strong.
These findings indicate that these designed marinades have shown excellent potential to improve food safety as well as shelf-life for the consumer, particularly in settings where food safety is often compromised such as barbecuing or in care settings. However, further recipe optimisation is required to make these marinades acceptable to consumers.
