2021, Food ChemistryCitation Excerpt :The main toxic substances retrieved in mushrooms are proteins/peptides able to promote cytotoxic effects acting on specific targets (Yin et al., 2019). Among these, can be included ribosome inactivating proteins (Wong et al., 2020), cytolysins as flammutoxin (Lin, Wu, & Shi, 1975), lectins able to destroy erythrocytes (Antonyuk, Klyuchivska, & Stoika, 2010) and also cyclic peptides as α-amanitin from Amanita phalloides, able to selectively inhibit RNA polymerase II (Meinecke & Meinecke-Tillmann, 1993). Recently, our research group has isolated and characterized from Agrocybe aegerita and Pleurotus ostreatus edible mushrooms novel fungal ribotoxin-like proteins (RL-Ps), enzymes that damage ribosomes cleaving a single phosphodiester bond within the universally conserved Sarcin-Ricin Loop (SRL) of ribosomes (Endo & Wool, 1982) releasing a characteristic RNA-fragment, known as α-fragment (Landi et al., 2017, 2020) and their findings, the first ever among those reported in the basidiomycetes, implemented the number of known enzymes that recognize the SRL region, a group that includes also ascomycetes ribotoxins (Olombrada et al., 2017) and ribosome inactivating proteins (Pizzo & Di Maro, 2016).
Combination of on-line desalting and HPLC-UV-ESI-MS for simultaneous detection and identification of FIP-fve and flammutoxin in Flammulina velutipes
2018, Journal of Food and Drug AnalysisCitation Excerpt :SDS-PAGE as well as UV-MS analysis provided corresponding molecular weight measurement for FIP-fve. Lin et al. (1974) [13] have isolated FTX with a molecular weight of 22 kDa from F. velutipes. Bernheimer and Oppenheim (1987) [14] have purified a hemolytic protein of 32 kDa from the same mushroom and assumed that the FTX of Lin et al. would have been proteolytically derived from their 32 kDa protein.
