Interleukin-1 signal transduction

Abstract

Interleukin-1 (IL-1) is primarily an inflammatory cytokine, although it is capable of mediating a wide variety of effects on many different cell types. Nearly every known signal transduction pathway has been reported to be activated in response to IL-1. However, the significance of many of these signaling events is unclear, due to the use of different and sometimes unique cell lines in studying IL-1-initiated signal transduction. Complicating matters further is the lack of association in many studies between identified IL-1-induced signals and subsequent biological responses. In this article, we review what is known about IL-1 receptor signaling and, whenever possible, correlate signaling events to biological responses.

      TNF-α is a cytokine, a cell signaling protein, involved in inflammatory responses. It is a potent activator of angiogenesis and activates fibroblast growth factor (FGF) and platelet activating factor (PAF) in endothelial cells (Bankers-Fulbright et al., 1996; Safi et al., 2018; Ryuto et al., 1996; Karar et al., 2021). It has been observed that TNF-α interacts with VEGF-A, FGF, and IL-1, to induce angiogenesis in endothelial cells (Yoshida et al., 1997; Defilippi et al., 1991; Safi et al., 2016a, 2017; 2016b;; Koolwijk et al., 1996).

      Administration of anti-TNFα monoclonal antibody ameliorates anemia and, in human TNFα transgenic mice, it reduces apoptotic erythroblasts [104]. Interleukin-1 (IL-1) is the first identified interleukin that plays a major role against infections and has been highlighted as a master regulator of inflammation, hematopoiesis and innate immunity [105]. It includes two related genes (IL-1a and IL-1b) which have different regulation but similar biological effects [106].
