2020, Clinics in Laboratory MedicineCitation Excerpt :Given the prevalence of diseases with genetic bases, the ability to edit genes will have an unparalleled impact on medicine and population health, and, as molecular diagnostics continue to improve, the targets for gene editing will only increase. The discovery of restriction enzymes was fundamental in the development of the recombinant DNA technologies that would become the cornerstone of gene therapy, including gene editing.3,4 Although ground breaking, the initial techniques lacked efficiency, with recombination only occurring in 1 of 103 to 109 cells5; specificity for site of recombination, with recombination happening at off-target sites at a high frequency6; and feasibility.
