The signal sequence of pre-proBNP is cleaved, resulting in the 108-amino acid proBNP (Sudoh et al., 1989). ProBNP is cleaved mainly by furin (an intracellular serine endopeptidase) (Nishikimi et al., 2015), producing the biologically active mature BNP (32 amino acids) and NT-proBNP (76 amino acids) when secreted into the bloodstream (Sudoh et al., 1989) (Fig. 1).

Human BNP is first synthesized as preproBNP (134 residues), after which its signal peptide is removed [93]. The resulting proBNP (108 residues) is then converted to bioactive BNP-32 and inactive NT-proBNP by selective cleavage at Arg76-Ser77 mainly by the type I transmembrane serine protease, furin [94–96].
