Silencing NAF-1 represses the proliferation and motility of PDAC cells through inhibiting HIF-1α

Abstract

Objective

To investigate the effect of NAF-1 on the proliferation and motility of PDAC cells and the molecular mechanism.

Methods

siRNA was used to knock down the expression of NAF1 in pancreatic cancer cells; qRT-PCR and western blot were performed to detect the level of NAF-1, HIF-1α, or EMT markers. The effects of NAF-1on cell proliferation and motility were assessed via colony formation, flow cytometry, wound healing assay, and transwell assays.

Results

NAF-1 was mainly localized in the cytoplasm according to the results of immunofluorescence. NAF-1 knockdown significantly repressed the proliferation and induced apoptosis of pancreatic cell. Besides, NAF-1 deficiency in Panc-1 cells obviously impaired the migration, invasion and EMT process. Mechanistically, HIF-1α mediates the biological function of NAF-1.

Conclusions

Silencing NAF-1 significantly represses the proliferation, inducing the apoptosis of pancreatic cell, besides, NAF-1 play a key role in repressing the motility of PDAC cells through inhibiting HIF-1α.
