Isolation and characterization of sex-linked SNP markers from transcriptomic sequences of the Chinese soft-shelled turtle (Pelodiscus sinensis)

Abstract

The Chinese soft-shelled turtle (Pelodiscus sinensis) exhibits sexual dimorphism in adults and the absence of sexually dimorphic morphological characteristics in the long-lived juvenile stage. The development of sex-linked SNP markers is of great significance for the improvement and replenishment of genetic resources. Here, we developed and characterized 35 novel polymorphic SNP markers based on the gonadal transcriptome of P. sinensis. The polymorphism information content, observed heterozygosity and expected heterozygosity of the 35 SNP markers ranged from 0.023 to 0.473, 0.012 to 0.547 and 0.023 to 0.503, respectively, and 10 of the SNPs significantly deviated from Hardy–Weinberg equilibrium (PHWE < 0.01). Two sex-linked SNPs were found to be significantly associated with sex phenotypes and showed 100% concordance across all tested individuals. These novel polymorphic SNP markers will be useful for future genetic studies of P. sinensis.
