Zusammenfassung
Hintergrund
Der nichtsyndromale Hörverlust ist die Erkrankung, für die beim Menschen bislang die häufigsten genetischen Veränderungen entdeckt wurden. Insgesamt wurden bisher 54 Loci für autosomal dominante, nichtsyndromale Hörstörungen identifiziert.
Patienten und Methoden
In dieser Studie untersuchten wir eine westfälische Familie mit einem progredienten tief- bis mittelfrequenten Hörverlust.
Ergebnisse
Eine genomweite Suche und Kopplungsanalyse bestätigten die Existenz eines neuen Locus (DFNA57). Der Phänotyp wurde auf einem 10-Mbp-Intervall auf dem Chromosom 19p13.2 von 7,8–18,2 Mbp kartiert. Für den Marker D19S586 wurde ein maximaler Zwei-Punkt-Lodscore von 3,08 erreicht. Die Region überlappt mit dem auf Chromosom 19 gelegenen Anteil des rezessiven Locus DNFB15.
Schlussfolgerung
Das Ergebnis unterstreicht, dass es sich bei den erblichen Hörstörungen um eine sehr heterogene Gruppe von Erkrankungen handelt. Die Identifikation der zugrunde liegenden Gene kann helfen, ein besseres Verständnis der molekularen Mechanismen des Hörens zu erreichen.
Abstract
Background
Non-syndromic hearing loss is the most genetically heterogeneous trait known in humans. To date, 54 loci for autosomal dominant non-syndromic sensorineural hearing loss (NSSHL) have been identified by linkage analysis.
Methods
In this study a German pedigree has been identified segregating a progressive bilateral loss of lower and middle frequencies.
Results
A genome-wide screening and linkage analysis revealed the existence of a new NSSHL locus (DFNA57). The phenotype was mapped to a 10°Mbp interval on chromosome 19p13.2 from 7.8 to 18.2°Mbp, a maximum 2-point LOD score of 3.08 was obtained for the marker D19S586. The region overlaps with the recessive locus DFNB15.
Conclusion
The results underline the heterogeneity of hereditary hearing disorders. Identification of genes can help to reach a better understanding of the molecular mechanism of hearing.
