Zusammenfassung
Aufgrund ihrer komplexen Beteiligung an der Ausbildung von Struktur- und Funktionsmerkmalen besitzen Fettsäuren eine zentrale Bedeutung unter den epidermalen Lipiden. Ihnen kommen als integrale Bestandteile von Zellmembranen, der interkorneozytären Barrierelipide und des oberflächlichen Hydro-Lipid-Filmes strukturbestimmende Eigenschaften und multiple Schutzfunktionen zu. Daneben fungieren endogene Fettsäuren als Mediatoren sowohl der epidermalen Zellproliferation und -differenzierung und damit der Lipidsynthese als auch inflammativer und immunologischer Reaktionen der Haut. Die topische Applikation ausgewählter Fettsäuren führt zu Wechselwirkungen auf struktureller und funktioneller Ebene. Sie kann einerseits Bedeutung für die Substitution einer gestörten Stratum-corneum-Barriere besitzen und andererseits die reversible Modulation der Barriereintegrität zum Zwecke eines optimierten Arzneistofftransportes in und durch die Haut bedingen. Mechanismen, die bei der Interaktion von applizierten Fettsäuren mit epidermalen Zellen, insbesondere Keratinozyten, eine Rolle spielen und Effekte, die die epidermale Proliferation und Differenzierung beeinflussen, sind das Anliegen aktueller Untersuchungen bzw. noch vielfach ungeklärt.
Summary
Fatty acids play a central role among the epidermal lipids because of their considerable contribution to structural and functional features of the epidermis. They are important components of cell membranes, the intercellular stratum corneum lipids and the hydro-lipid skin surface film. Additionally, endogenous fatty acids act as mediators of the epidermal cell proliferation and differentiation and therefore, lipid synthesis and also as mediators of inflammatory and immunological processes.
The topical application of certain fatty acids leads to interactions at the structural and functional level. On the one hand, fatty acids may be of importance to restore a disturbed stratum corneum barrier. On the other hand, fatty acids are able to modulate dermal and transdermal drug transport. Mechanisms of the interaction between exogenous fatty acids and epidermal cells, especially keratinocytes, and effects on their proliferation and differentiation are currently under investigation or largely unknown, respectively.
