Summary
To elucidate the post-mortem proteolysis we investigated the behaviour of the free amino acids in rotting liver homogenates by means of two-dimensional thin-layer chromatography and reflection densitometry. The post-mortem alterations of the amino acid levels are characterized mostly by a two-phase increase with maxima at the end of the first and after the third week. A few amino acids showed only little changes hardly above the physiological intra-cellular levels. Anaerobic rot experiments and experiences with antibiotic treatment give rise to the supposition that increasing amino acid concentrations indicate a predominance of proteolytic activities over amino acid catabolism. In the beginning autoenzymes are prevalent, while afterwards bacterial proteases probably predominate. The transient regression about the second and third week is interpreted as the effect of a temporarily intensified amino acid degradation by foreign enzymes (maximal bacterial growth).
Zusammenfassung
Als Beitrag zur Aufklärung des postmortalen Eiweißabbaus wurde das Verhalten der freien Aminosäuren in faulendem Leberbrei mittels zweidimensionaler Dünnschichtchromatographie und Reflexionsdensitometrie untersucht. Die postmortale Entwicklung der Aminosäurespiegel ist in der Regel durch einen zweiphasigen Anstieg mit Gipfeln etwa zum Ende der ersten und nach der dritten Woche gekennzeichnet; in einigen Fällen fanden sich auch nur geringe Bewegungen kaum oberhalb des physiologischen Intrazellular-Spiegels. Anaerobe Faulversuche und das Verhalten nach Zusatz eines Antibiotikums lassen vermuten, daß die Konzentrationserhöhungen ein Überwiegen der Proteolyse gegenüber dem Aminosäureabbau anzeigen, wobei anfangs Eigenfermente dominieren, während später wahrscheinlich bakterielle Proteasen überwiegen. Der vorübergehende Rückgang der Aminosäurekonzentrationen um die zweite bis dritte Woche wird als Folge eines kurzzeitig forcierten Aminosäureabbaus durch Fremdenzyme (Maximum des Bakterienwachstums) angesprochen. Die Konzentrationsveränderungen sind temperaturabhängig. Aus den Ergebnissen von Brut- und Kühlschrank-Versuchen ergibt sich ein Q10 von etwa 2 bis 3.
