Two nervous pathways are part of the light-sensitive pineal complex in Rana esculenta (Dodt et al.): the frontal organ, which is situated in the skin, and from which the pineal nerve originates; and the intracranial epiphysis cerebri, from which the pineal tract takes its origin. Both tracts are afferent, i.e. directed towards the central nervous system. Signs of regressive development are found in the frontal organ of adult R. esculenta. The same phenomenon can be observed in the pineal nerve which is the nervous connexion between frontal organ and epiphysis cerebri.

The number of fibres that form the nervus and tractus pinealis, and their components, can only be determined by means of electron micrographs. The determination of the number of fibres has also been done after the electric activity of the nervus and tractus pinealis had been registered.

Nervus and tractus pinealis contain unmyelinated as well as myelinated elements. The unmyelinated nerve fibres outnumber the myelinated ones. The same relation is observed in the fibre components of the nervus opticus (Maturana 1960). The pineal nerve is rich in connective tissue sheaths.

One to 22 myelinated and 35–146 unmyelinated nerve fibres are counted in the pineal nerve of Rana esculenta. Fixed myelinated fibres have a diameter of 2–7 μ (thickness of the myelin sheath: 0.2–0.6 μ), whereas unmyelinated fibres have a diameter of 0.2–0.8 μ (mostly 0.3–0.5 μ).

The pineal tract of Rana esculenta shows 60–80 myelinated and up to 200 unmyelinated fibres. Fixed myelinated fibres have a diameter of 1–6 μ (thickness of the myelin sheath 0.1–0.4 μ); the diameter of the unmyelinated fibres is 0.1–1.0 μ.

In spite of the new results obtained by electron microscopy, no definite statements are possible as to which of the nervous structures is responsible for the chromatic and achromatic responses of the frontal organ.

Some unmyelinated fibres in the pineal tract display granules, which are about 850 to 950 Å in size. These granules have a sheath and an electron-dense internal structure. They resemble inclusions in nerve fibres, which contain biogenic amines. It is not known whether these peculiar fibrous elements in the pineal tract are afferent or efferent.

The perivascular boundary layer of the epiphyseal parenchyma shows similar profiles, which are filled with granules up to 0.2 μ in size.

Synaptic structures are seen in the region between the pineal tract and the basis of the subcommissural organ. The origin of the elements, involved in these contacts, is unknown. Unusually wide extracellular spaces, which cannot be regarded as artefacts, are also seen in this region.

Ranvier's nodes are sometimes found along the pineal tract. Dilated extracellular spaces extend to that part of the axon, that is free of sheaths. Ranvier's nodes are also seen in the pineal nerve.

Zwei Nervenbahnen sind ein Teil des lichtempfindlichen (Dodt u. Mitarb.) Pinealkomplexes von Rana esculenta. Das in der Haut gelegene Stirnorgan (Frontalorgan, Epiphysenendblase) entsendet den Nervus pinealis (Tractus frontalis), die intrakraniale Epiphysis cerebri — den Tractus pinealis (Tractus epiphyseos). Beide Bahnen sind afferent, auf das nervöse Zentralorgan gerichtet. Am Stirnorgan adulter Esculenten finden sich Zeichen einer regressiven Entwicklung. Diese erfaßt auch die nervöse Verbindung des Stirnorgans mit der Epiphysis cerebri — den Nervus pinealis.

Nur elektronenmikroskopische Aufnahmen erlauben eine genaue Auskunft über die Faserzahl und Faserzusammensetzung des Nervus und Tractus pinealis. Die Bestimmung der Faserzahl im Nervus und Tractus pinealis wurde auch nach vorheriger Registrierung ihrer elektrischen Aktivität durchgeführt.

Nervus und Tractus pinealis enthalten sowohl markhaltige als auch marklose Elemente. Die Zahl der marklosen Nervenfasern überwiegt. In dieser Relation ist eine Parallele zur Faserarchitektonik des Nervus opticus (Maturana 1960) sichtbar. Der Nervus pinealis ist sehr reich an bindegewebigen Hüllstrukturen.

Im Nervus pinealis von Rana esculenta wurden 1 bis 22 markhaltige und 35 bis 146 marklose Nervenfasern gezählt. Die markhaltigen haben im fixierten Zustand einen Durchmesser von etwa 2–7 μ (Markscheidendicke etwa 0,2–0,6 μ), die marklosen — von 0,2 bis 0,8 μ (meist 0,3 bis 0,5 μ).

Im Tractus pinealis von Rana esculenta wurden 60 bis 80 markhaltige und bis 200 marklose Nervenfasern gezählt. Die markhaltigen haben im fixierten Zustand einen Durchmesser von 1 bis 6 μ (Markscheidendicke 0,1 bis 0,4 μ), die marklosen — 0,1 bis 1,0 μ.

Zur Frage der für die chromatische und achromatische Antwort des Stirnorgans (Dodt u.a.) verantwortlichen nervösen Strukturen können — ungeachtet der neuen elektronenmikroskopischen Einblicke — noch keine beweiskräftigen Angaben gemacht werden.

Der Tractus pinealis führt vereinzelte marklose Fasern, die mit 850–950 Å großen Granula gefüllt sind. Diese Granula haben eine Hüllmembran und eine elektronendichte Innenstruktur. Sie erinnern sehr stark an die Einschlüsse von Nervenfasern, deren Gehalt an biogenen Aminen gesichert ist. Der afferente oder efferente Charakter dieser eigenartigen Faserelemente des Tractus pinealis ist unbekannt.

Mit ähnlichen, z.T. sogar bis 0,2 μ großen Granula gefüllte Profile beobachtet man auch an den zerklüfteten perivasculären Grenzflächen des Epiphysenparenchyms.

In der Zone zwischen dem Tractus pinealis und der Basis des Subcommissuralorgans wurden synapsenähnliche Strukturen nachgewiesen. Die Herkunft der an diesen Kontakten beteiligten Elemente ist noch nicht geklärt. Hier finden sich außerdem überdurchschnittlich weite extrazelluläre Räume, die nicht als Artefakte angesehen werden können.

Im Verlauf des Tractus pinealis können Ranviersche Schnürringe auftreten. Erweiterte Extrazellularräume erstrecken sich bis an den hüllenfreien Abschnitt des Axons. Ranviersche Schnürringe kommen auch im Nervus pinealis vor.
