Minimal Cells and Genome Minimization: Top-Down and Bottom-Up Approaches to Construct Synthetic Cells

Abstract

Bioproduction of high-value molecules has traditionally been carried out using engineered host cells. However, toxic molecules and membrane proteins are difficult to produce using this approach. Minimal cells, which are synthetic enclosed compartments that mimic the cell, are being explored as an alternative to live cells. Apart from catalysing enzymatic reactions, minimal cells are being constructed that can maintain homeostasis and are also capable of evolution, though in a limited sense. This bottom-up approach is complemented by research in genome minimization, a top-down approach to converting cells into dedicated biofactories. This chapter summarizes studies pertaining to genome minimization and construction of minimal synthetic cells.
