Abstract

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) is an analytical technique for fast and precise mass determination of biological molecules. Intact molecular ions are produced by short pulsed laser irradiation of the biomolecules which are enbedded in a matrix consisting of small highly absorbing organic molecules. Mass analysis is carried out in a linear or reflector time-of-flight mass spectrometer. The accessible mass range is 500 000 Da, a mass accuracy of up to 0.01% can be reached, the sample amounts required are 1 pmol or less. Proteins, glycoproteins, oligonucleotides and oligosaccharides can be analyzed.
