Abstract
Immunization with early, impure rabies vaccine developed from infected central nervous system (CNS) tissue produced encephalomyelitis in some humans (reviewed in Hemachudha et al. 1987). Based on this observation, several investigators later showed that immunization of animals with heterologous extracts of brain tissue could produce inflammatory lesions in the CNS white matter accompanied by demyelination (reviewed in Weigle 1980). In 1947, four investigators independently reported the induction of encephalomyelitis in guinea pigs, monkeys and rabbits using one injection of CNS tissue emulsified in complete Freund’s adjuvant (CFA). However, no one was able to induce the disease in mice until Bernard and Carnegie immunized SJL/J mice with pertussis vaccine in addition to spinal cord homogenate or myelin basic protein (MBP) emulsified in CFA (Bernard and Carnegie 1975). Another protein component of myelin, myelin proteolipid apoprotein (PLP) (SATOH et al. 1987; TUOHY et al. 1988) has since been shown to be encephalitogenic.
