The Study of Comparing the Efficiency of ZnPc-PDT and HPD-PDT in Killing Mice Lewis Lung Cancer Cells

Abstract

Photodynamic therapy(PDT) has been developed as an alternative therapy of cancer, but at present PDT for lung cancer is still using Hematoporphyrin Derivative (HPD). The aim of this study is to compare the efficiency of two photosensitizers ZnPc and HPD in killing lewis lung cancer(LLC) cells under different concentrations, exposure time and power density, and explore the best parameters of ZnPc-PDT for lung cancer. This research selected the second generation photosensitizer ZnPc and traditional photosensitizer HPD as the research objects, applied them to the LLC lung cancer cells, used spectrophotometer to measure the absorption peak values and selected proper excitation light resources; using inverted microscope, MTT, FACS to observe and compare the killing effects of ZnPc-PDT and HPD-PDT on LLC Lewis lung cancer cells under different photosensitizers’ concentration, exposure time, and different laser power density. The results of our study suggest that comparing to HPD, ZnPc has a singular absorption wave peak with longer wavelength; In both cases, the effects of killing lung cancer increases with the increasing of photosensitizers’ concentration, exposure time, and different light source power density, while the effects would be saturated after reaches certain value; At the same conditions, ZnPc is more effective in killing lung cancer than HPD with distinctive difference. These findings shows that ZnPc-PDT is a more effective treatment in curing lung cancer than HPD-PDT. This investigation provides valuable information in making the choice of photosensitizers as well as relevant dosage usage in body experiments and last phase clinical applications.
