ارزیابی واکسن تهیه‌شده از باکتری کشته‌شده Vibrio fluvialis در مرغ تخم‌گذار نژاد هایلاین W36

چکیده

باکتری Vibrio fluvialis موجب بیماری در ماهیان پرورشی و ضرر‌های اقتصادی قابل توجه می‌شود. این باکتری پاتوژن انسانی و چندین نوع از ماهیان پرورشی دریایی می‌باشد. استفاده از تکنولوژی ایمونوگلوبولین زرده تخم‌مرغ (IgY) تا کنون برای کنترل عفونت‌های میکروبی فراوانی مورد استفاده قرار گرفته است. هدف از مطالعه حاضر بررسی و ارزیابی واکسن تهیه شده از باکتری V. fluvialis  پس از تزریق به مرغ تخم‌گذار می‌باشد. باکتری V. fluvialis پس از کشت در محیط مایع، با فرمالئین 05/0 درصد مجاور شد. سپس سلول باکتری به همراه اجوانت فروند به عنوان واکسن به مرغ‌های تخم‌گذار نژاد هایلاین در 7 نوبت تزریق شد، افزایش وزن مرغ‌ها تا بلوغ جسمی ‌و تولید تخم‌مرغ مورد ارزیابی قرار گرفت، سپس آزمایش الایزا غیر مستقیم برای شناسایی IgG ضد باکتری V. fluvialis  طراحی و اجرا شد. تزریق واکسن تهیه شده در افزایش وزن مرغ‌ها در طول آزمایش و تعداد و وزن تخم‌مرغ‌ها معنی‌دار نبود و آزمایش الایزا نشان داد تیتر آنتی‌بادی در سرم مرغ‌های هایپر ایمن شده پس از تزریق چهارمین بوستر به 1:12800 رسیده است. با توجه به نتایج به دست آمده و نتایج مطالعات سایر محققین به نظر می‌رسد، آنتی‌بادی زرده تخم‌مرغ (IgY) که همان IgG منتقل شده به زرده است، توانایی مهار بیماری‌زایی این باکتری را در حیوان آزمایشگاهی و مزرعه دارد.

Vibrio fluvialis causes disease and significant economic losses in breeding fish. This bacterium is considered as a human pathogen and several species of marine spawning fish. The egg yolk immunoglobulin technology (IgY) has been used to control many microbial infections. The purpose of the present study was to evaluate the vaccine prepared from V. fluivialis bacteria after injection into laying hens. V. fluvialis was admixed by 0.05% formaldehyde after culture in a liquid medium. Then, the pathogen along with Freund’s adjuvant was injected to the Hyline W36 hens as a vaccine for 7 days. Increase of the weight of the hens was measured until Physical maturity and egg production. Then, an Indirect ELISA was designed to detect IgG Anti V. fluivialis. Injection of the vaccine had no significant effect on weight gain of hens and the number and weight of eggs during the experiment. ELISA test showed that antibody titers in the serum of hyper-immunized hens after injection of the fourth booster reached 1: 12800. According to the results obtained and the results of studies by other researchers, egg yolk antibody (IgY), which is the same as IgG, has been shown to inhibit the pathogenicity in lab and farm animals.
