Abstract: Im Mittelpunkt dieser Arbeit standen die Untersuchung der Verbreitung von Calysteginen in verschiedenen Pflanzenfamilien und biosynthetischer Schritte in Cochlearia officinalis L. (Löffelkraut). In den Familien Moraceae, Erythroxylaceae und Brassicaceae wurde nach Calysteginen gescreent. In allen Familien wurden Calystegine gefunden. Cochlearia officinalis L. enthält sowohl Calystegine als auch Tropinester (z.B. Cochlearin). Es gelang die Amplifikation eines tr-homologen Fragmentes mit Hilfe von RT-PCR und degenerierten Primern. Das Fragment wurde mit RACE-PCR vervollständigt. Die resultierende cDNA-Sequenz weist alle Merkmale einer kurzkettigen Dehydro-genase/Reduktase (SDR) auf. Der Vergleich mit Tropinonreduktasen (TR) aus Solanaceae zeigt auf Aminosäureebene eine durchschnittlich 57%-ige Identität. Das Enzym wurde in E. coli rekombinant exprimiert (6xHis-tag) und affinitätschromatographisch gereinigt. Es katalysiert sowohl die Reduktion von Tropinon zu Tropin und Pseudotropin als auch die Oxidation von Tropin und Pseudotropin. Die Affinität zu Tropinon ist geringer als die der TR von Solanaceae. Ein molekulares Modell des Enzyms wurde auf der Basis der kristallisierten TRII von Datura stramonium L. erstellt und die Substratorientierung durch Dockingversuche untersucht. Die Erkenntnisse aus dem Docking wurden durch Mutageneseversuche bestätigt. Zu Vergleichszwecken wurde parallel eine der 15 "putativen" TR aus Arabidopsis thaliana L. mit identischen Methoden untersucht. Durch PCR-basiertes Screening einer cDNA-Bank von Cochlearia officinalis L. wurde ein vollständiger spds-homologer cDNA-Klon isoliert. Zur Bestätigung der biochemischen Funktion wurde die Sequenz in E. coli als His-tag-Fusionsprotein exprimiert. Das resultierende Protein war löslich und zeigte keine spezifische Aktivität, die gefundene SPDS-Aktivität war nicht reproduzierbar und eine enzymkinetische Auswertung nicht möglich. Zusätzlich wurde mit dem PMT-Cosubstrat SAM wiederholt ein Produkt gefunden, das in UV-Spektrum und Retentionszeit N-Methylputrescin entspricht.
The main focus of this thesis was to analyse the distribution of calystegines (polyhydroxy-nortropane alkaloids) within plant families and to investigate the biosynthetical steps in Cochlearia officinalis L. Moraceae, Erythroxylacae and Brassicaceae were screened for calystegines. Species from all three families contained calystegines. Cochlearia officinalis L. contains calystegines as well as tropane esters (e.g. cochlearine). A tropinone reductase homologuos fragment was cloned by RT-PCR with degenerated primers. The full length clone was obtained by RACE-PCR and exhibits the characteristic features of short chain reductases/dehydrogenases. When compared to tropinone reductases (TR) from Solanaceae identity averages about 57% on amino acid level. The enzyme was heterologuously expressed in E. coli (6xhis-tag) purified by affinity chromatography. It catalyses the reduction of tropinone to both stereoisomers as well es the oxidation of both tropinols (tropin/pseudotropin). The affinity for tropinon is lower than in Solanaceae TR. Based on the crystal structure of Datura stramonium TR II a molecular model of the enzyme was developed and used for docking studies where substrate orientation was investigated. Findings from docking studies were confirmed by site directed mutagenesis experiments. As comparative means 1 of 15 "putative" TR from Arabidopsis thaliana L. was examined accordingly. By PCR-based screening a spermidin synthase (SPDS) homologuous clone was found and subsequently expressed in E. coli. The resulting protein was soluble but showed no specific activity. SPDS activity was found to be nonreproducible. An additional product resembling Nmethylputrescine in retention time and UV-spectrum was found with cosubstrates dcSAM and SAM, respectively.
