Destruction, oxidation of DNA and lipids of mitochondrial membranes in chronic viral hepatites C, B

The objective. To establish the degree of DNA damage of lymphocytes, liver cells and to determine the content of DNA damage markers, products of lipid peroxidation and antioxidant system parameters in patients with chronic viral hepatites B (CHB) and C (CHC).

Materials and methods. Autopsy materials of 31 patients with CHC for the period 2012–2017 were studied. The control group comprised autopsies of 16 patients, who died of noninfectious diseases. The subjects were divided into groups according to the METAVIR liver fibrosis stages. We determined: DNA damage by the comet assay, levels of 8-hydroxy-2-deoxyguanosine (8-OHdG), 8-nitroguanine (8-NO2G), total antioxidant capacity (TAC), reduced glutathione (GSH), malondialdehyde (MDA) and 4-hydroxy-2,3-nonenal (4-HNЕ). Statistical processing used the STATISTICA 10.0 package.

Results. The level of DNA damage in patients with CHC and CHB is 2 times higher as compared with the norm. In viral hepatites, changes in the liver increase smoothly from stage to stage, correlating with the increase of 8-OHdG, 8-NO2G levels. At advanced stages of fibrosis F3-F4, MDA and 4-HNE serum concentrations increased in all groups, but in the liver this picture is observed as soon as at the stage F1. Regular specificities reflecting depletion of TAC and GSH with the increasing intensity of lipid and DNA oxidation have been found.

Conclusion. DNA toxicity, increased concentrations of DNA, lipid membrane destruction markers and proportional depletion of antioxidant reserves with the development of liver fibrosis have been shown.
