Abstract:
Regeneration potential of isolated organs of Daendrobium antennathum and D. phalaenopsis has been investigated in vitro, using varies combinations of agar-agar solidified RM medium at pH 5.2. Growth regulators added to medium, especially cytokinin (BA) stimulated the development of lateral buds on isolated shoots of the two examined species, whereas stimulating effect of peptone was distinct in D. antennathum only.
Auxins delimited the development of shoots, but stimulated root formation.
In D. phalaenopsis the callus was formed at the base of isolated shoots, especially in presence of BA alone and in its interaction with peptone or auxins.
The development of callus which gave rise to protocorms allows a clonal mass propagation of D. phalaenopsis by means of the above method.
