PHLOX PLANTS FROM SHOOT TIPS

Abstract:
Shoot tips (1 mm), containing the meristem and 1 to 2 pairs of leaves, were isolated in July from Phlox paniculata 'Orange' stock plants grown in the field. For normal shoot and root development there was no need for exogenous suppleyed IAA. With increasing concentration of IAA there was inhibition of both shoot and root development. NAA was much stronger than IAA in its inhibitory effect on shoot growth, but roots did grow from the developed callus at high NAA concentrations. Kinetin was necessary for good development of shoots and roots.

There was found a relationship between root development and shoot length. Plants with 1 and 2 roots had a shoot length at 22 mm or more, and plants without roots had an average shoot length of only 11 mm. This indicates that at a shoot length of 22 mm or more, the shoot is able to synthesise its own hormones in the proper concentration for the initiation of roots.
