한방생약자원으로부터 천연항산화제를 개발하기 위한 연구의 일환으로 남천 열매 추출물에 대한 폴리페놀과 플라보노이드 화합물 함량, 그리고 항산화 활성을 측정하였다.
폴리페놀 화합물 함량은 물을 용매로 추출한 RW에서115.06 mg/g을 함유하였으며, 플라보노이드 화합물은 에탄올을 용매로 추출한 RE에서 60.73 mg/g을 함유하였다. Free radical에 대한 전자공여 효과를 측정한 결과, 물과 에탄올추출물 모두 0.1 mg/mL의 농도에서 80% 이상의 활성을 나타내었으나 남천 열매 추출물의 농도 증가에 따른 유의적차이가 없었다. SOD 유사활성능은 1.0 mg/mL의 농도에서RW 42.88%, RE 45.96%의 활성을 나타내었다. 아질산염 소거능은 에탄올 추출물인 RE pH 1.2조건의 1.0 mg/mL에서60.92%였으며, pH 3.0에서는 62.70%의 소거효과를 나타내었다. Xanthine oxidase에 대한 저해 효과를 측정한 결과에서는 1.0 mg/mL의 농도에서 남천 열매 물 추출물인 RW 96.97%로 RE (57.71%)보다 약 1.8배 우수한 저해효과를 나타내었다. 이상의 결과, 민간에서 약용으로 사용되는 남천열매는 우수한 항산화 활성을 지닌 천연소재임을 확인 하였으므로 이를 항산화 활성을 지닌 미용제품 및 기능성 제품의 원료 및 첨가재로 활용할 수 있을 것이라 사료된다.
This study was carried out to investigate the antioxidative effects of water and ethanol extracts from the fruit of Nandina domestica as part of the development of functional beauty materials. The highest content of polyphenol compound was 115.06 mg/g in the water extract, and the content of flavonoid compounds was the highest as 60.73 mg/g in the ethanol extract. In the measurements of electron donating ability, both extracts revealed high abilities more than 80% at 0.1 mg/mL concentration. The SOD-like activities of water and ethanol extracts showed the highest effect as 42.88% and 45.96% at 1.0 mg/mL concentration, respectively. In the analysis of nitrate scavenging activity, ethanol extract showed the highest activity as 60.92% and 62.70% in the condition of pH 1.2 and pH 3.0 at 1.0 mg/mL concentration, respectively. The xanthine oxidase inhibitory rate of water extract had the highest effect as 96.97% at 1.0mg/mL concentration. These results verified that water and ethanol extracts from the fruit of N. domestica have a strong antioxidant activity and can be used as an effective antioxidant source for manufactures, medicines and cosmetic stuffs.
