Abstract

A method is described for the estimation of the pigment content in chloroplasts by means of HPLC. The method is highly sensitive. Less than one picomole of individual pigments - equivalent to 1 milligramme fresh weight of spinach leaves - is needed for quantitative analysis. Analysis time is only 15 min, whereas in routine determination by thin layer chromatography a single sample needs 4 hours to be completed.

Moreover the pigments are protected against desintegration by light and air. Using a isocratic HPLC -method no equilibration of coloumns is necessary, thus permitting four runs per hour.
