본 연구는 self-motile이라는 독특한 특성을 가진 MnO₂를 도핑한 규조류 복합체를 과산화수소와 반응시켰을 때 나타나는 active micro-locomotion을 이용하여, 의치 및 치과용 구내 장치에 대한 세정효과를 식품의약품안전처의 의치세정제 세척력 효력평가법 가이드라인에 따라 시판 의치세정제와 비교하여 알아보고자 하였다. 미국치과의사협회 제시 기준에 따라 인공타액을 제조하여 96웰플레이트에 분주, 건조시킨 후 양성대조군(10~15% NaOCl), 음성대조군(증류수), 다양한 조건의 실험군들(규조류 복합체 A (Aulacoseira, MnO₂-polydopamine (PDA)-A), 규조류 복합체 M (Melosira nummuloides, MnO₂-sugar(S)-M), 폴리덴트(GlaxoSmithKline, Dungarvan, Ireland), 덴트픽스-포르테(Helago-Phama GmbH&Co., Parchim, Germany)들과 반응시켰다. 세척 후 흡광도(OD 600)를 측정하여 70% 이상이면 ‘세척력 효력’이 있는 것으로 판정하고, 세정 효과의 군간 비교를 위해 one-way ANOVA, Bonferroni correction을 시행하여 통계적 유의성을 검정하였다(p<0.05). 그 결과 모든 군에서 ‘세척력 효력’이 있는 것으로 판명되었으며, 평균 세척률은 양성대조군 NaOCl에서 93.8±1.0%, 음성대조군 증류수에서 79.1±1.5%, 규조류 복합체 A군은 3% H₂O₂ 2 mg/mL과 반응 시 79.8±4.5%, 6% H₂O₂ 2 mg/mL에서 64.7±5.5%, 6% H₂O₂ 4 mg/mL에서 81.9±7.9%의 평균 세척률을 보였다. 규조류 복합체 M군은 3% H₂O₂ 2 mg/mL에서 88.5±3.6%, 6% H₂O₂ 2 mg/mL에서 75.8±4.0%, 6% H₂O₂보였다. 양성대조군은 모든 실험군과 유의한 차이가 있었으나, 음성대조군은 A2와 A3, M1과 M2, M3와, 폴리덴트, 덴트픽스-포르테에서만 유의한 차이를 보였다(F=190.141, p<0.001). 양성대조군을 제외한 모든 군 중에서 MnO₂-S-M을 3% H₂O₂ 2 mg/mL의 비율로 반응시켰을 때 세정효과가 가장 좋았고, 규조류 복합체의 active micro-locomotion을 이용한 세정 효과가 시판중인 의치 세정제에 상응하는 결과를 보이므로 세척력 효력이 있는 것으로 평가되며, 보다 높은 세정 효과를 위한 최적의 적용 조건은 추가적인 연구가 필요할 것으로 사료된다.
This study compared the effectiveness of self-propelling diatom microbubblers to clean dental appliances with commercial denture cleaning agents according to the Ministry of Food and Drug Safety’s guidelines. The microbubbler is made by doping diatoms with MnO₂ nanosheets that can decompose hydrogen peroxide to generate oxygen bubbles. Artificial saliva is prepared in accordance with the criteria presented by the American Dental Association, dispensed, and dried in 96 well plates. Experimental groups include 10-15% NaOCl (positive control), distilled water (negative control), diatom microbubbler A (Aulacoseira, MnO₂-polydopamine (PDA)-A), diatom microbubbler M (Melosira nummuloids, MnO₂-sugar (S)-M), Polident (GlaxoSmithKline, Dungarvan, Ireland), Dentfix-forte (Helago-Phama GmbH&Co, Parchim, Germany). After washing, absorbance (OD 600) was measured. If the absorbance was 70% or higher, the condition was determined to have “cleaning power potency,”Statistical significance was evaluated by one-way ANOVA and Bonferroni correction to compare cleaning effects among groups (p<0.05). The average cleaning rates were 93.8±1.0% in NaOCl (positive control) and 79.1±1.5% in distilled water (negative control). With the diatom microbubbler A, the average cleaning rate was 79.8±4.5% in the 3% H₂O₂ 2 mg/mL, 64.7±5.5% in 6% H₂O₂ 2 mg/mL, and 81.9±7.9% in 6% H₂O₂ 4 mg/mL. The diatom microbubbler M group showed average cleaning rates of 88.5±3.6% in 3% H₂O₂ 2 mg/mL, 75.8±4.0% in 6% H₂O₂ 2 mg/mL, and 84.5±4.5% in 6% H₂O₂ 4 mg/mL. Finally, conventional denture cleaning agents showed average cleaning rates of 88.2±1.2% in Polident and 83.3±3.0% in Dentfix-forte. The positive control group had significant differences from all experimental groups, but the negative control group showed significant differences only in A2 and A3, M1 and M2, M3, Polident, and Dentfix-forte (F=190.141, p<0.001). Among all groups except the positive control group, MnO₂-S-M mixed with 3% H₂O₂ 2 mg/mL showed the highest cleaning rate. As the results of this study show, diatom complexes exhibit cleaning effects compatible with conventional denture cleaning agents. Further studies need to be conducted to narrow down the specific optimal conditions of diatom microbubblers and maximize the cleaning effect.
