In Vitro Cultivation of Nodal Segments in Quercus mongolicaChinese Full Text

Abstract: Strong nodal segments of aquiculture were used as a source of initial explants to study the effects of culture media and hormones on organogenesis and plantlet regeneration of Quercus mongolica in vitro.Results showed that the media containing 0.2 g·L-1 PVP could effectively inhibit stem browning.The 1/2MS media with 0.2 mg·L-1 6-BA were more suitable for initial culture,and new shoots started early,with an average height of sprouts of 3.00 cm and the highest one of 5.20 cm.1/2MS medium supplemented with 0.15 mg·L-1 6-BA and 0.15 mg·L-1 KT in subculture was the best,with an average height of sprouts of 3.00 cm and an average multiplication coefficient of 3.33.High concentration of 6-BA was harmful to shoot growth due to the fact that the base of shoots was completely filled with callus tissues and vitrification appeared.High rooting percentages and excellent development root systems were obtained on MS medium with NAA 0.02 mg·L-1 and IBA 0.05 mg·L-1,with a rooting rate of 62.5%.
