Inhibition of Candida parapsilosis Fatty Acid Synthase (Fas2) Induces Mitochondrial Cell Death in Serum

Cell death is dependent on mtDNA.

A, mitochondrial DNA and genomic DNA were evaluated by PCR from total DNA isolated from the indicated strains. Mitochondrial DNA content was assessed using the subunit I of cytochrome c oxidase gene (Cox1). Genomic DNA content was determined using the actin gene (Actin). B, Deletion of mtDNA protected Fas2Δ/Δ yeast cells from death induced by culture in serum. WT, Fas2Δ/Δ and Fas2Δ/Δ with partially deleted mtDNA (Fas2-rho) were incubated with 50% serum for 24 and 48 h. Aliquots were serially diluted and WT yeast were spotted onto YPD whereas the Fas2Δ/Δ mutants were inoculated onto YPDT40. Experiments were performed twice with similar results. C, A proposed model of yeast cell sensitivity to glucose in the media in the setting of fatty acid inhibition.
