Induction of Eosinophil Apoptosis by the Cyclin-Dependent Kinase Inhibitor AT7519 Promotes the Resolution of Eosinophil-Dominant Allergic Inflammation

AT7519 effects on eosinophil apoptosis and subsequent clearance by macrophages is caspase dependent.

(A) Schematic representation of the experimental protocol. Immunized mice were challenged with OVA and 24 h later were treated with AT7519 and/or zVAD-fmk. The percentage of apoptotic eosinophils (B) and percentage of macrophages containing apoptotic bodies (C) was assessed 30 h after antigen-challenge. Results are expressed as the % cells per cavity, as a mean ± SEM of at least five mice in each group. ***P<0.001 when compared with vehicle-treated, OVA-injected mice. #P<0.05, ### P<0.001 when compared with AT7519-treated, OVA-injected mice.
