Enzymatic hydrolysis of silk fibroin for production of α-glucosidase inhibitorsChinese Full Text

Abstract: Silk fibroin was hydrolyzed by alcalase to produce the peptide with high α-glucosidase inhibition activity in vitro.The hydrolysis condictions were optimized according three indicators through orthogonal tests and the optimum reaction conditions were determined as follows: pH8.5,temperature 60 ℃,substrate concentration 5%,enzyme dosage 1200 U/g protein,and reaction time 100 min,under which the α-glucosidase-inhibition rate and DH of the product were 44.90% and 16.80%,respectively.The results shows that the α-glucosidase inhibition activity in vitro was related to the concentration of silk fibroin and the reaction time could be 40 min.Moreover,microamounts of glucose existing in silk fibroin,but they had less interrupting effect on the anti-hyperglycemic activity.
