Optimization of fermentation medium components for Bacillus FC96 by response surface analysisChinese Full Text

Abstract: In order to improve the biomass of Bacillus FC96 liquid medium,Single factor test to determine the optimum carbon and nitrogen sources,inorganic salts are glucose,beef extract,potassium dihydrogen phosphate.Using response surface analysis to determine the optimum ratio of these three substances.The results showed that best medium composition of Bacillus FC96 are glucose 12.11 g/L,beef extract 23.31 g/L,and potassium dihydrogen phosphate 2.33 g/L.According to the theoretical model predicted the maximum bacteria number of 2.85×109 cfu/mL.In the initial experimental conditions,bacteria number was 2.32×109 cfu/mL.In the optimization of the best medium for proliferation of the culture of Bacillus FC96 viable cells obtained was 2.97×109 cfu/mL,bacteria ratio of 28% before optimization,the error of test values and predicted values is 4.21%.
