Response surface methodology to optimize the extraction of aflatoxin B1 from koji beanChinese Full Text

Abstract: Bean used in Pixian soybean paste are produced in Sichuan province and Yunnan province,koji bean are produced by screening,dipping,vaccination and a series of process.In this paper,according to the box-behnken desigh software,we used the enzyme-linked immuneosorbent assay(ELISA) to test the valve of aflatoxin B1 in koji bean.Taking over the influence about methanol concentration,the amount of methanol solution and ultrasonic time on the extraction of aflatoxin in the samples to determine the best proposal.In addition,seting the aflatoxin content as response value,and using response surface methodology to optimize the extraction parameters.Results show that: if methanol concentration is 39.66%,ultrasonic time is 27.70 min,ultrasonic power is 88.97%,by this optimized condition,detecting starter propagation of broad beans can achieve its maximum value for 1.47 μg/kg.
