Abstract: Directed evolution has been proven to be an effective method for proteins engineering. In the past few years, many new protocols for directed evolution were developed on the basis of oligonucleotide -directed random mutation, error-prone PCR and DNA shuffling. These new methods and their characteristics were summarized to provide a reference for choosing an approach to modify specific protein properties. The recent researches indicated that the combined directed evolution, coupling the directed evolution with rational design and computational design approaches respectively, is becoming increasingly important in protein engineering and is becoming the new direction for directed evolution.
