Establishment of regeneration system of Hemeroallis middendorfii Trautv.et Mey ‘Sweet Treasure’Chinese Full Text

Abstract: Hemeroallis middendorfii Trautv.et Mey‘Sweet treasure’was used as explants to study the effect of different hormones and their concentrations on the callus induction,proliferation,adventitious bud and roots induction by orthogonal experiment.The results showed that 6-BA was a key factor on the callus induction,the effect sequence of four hormones was 6-BA＞2,4-D＞NAA＞KT,the optimum callus induction medium was MS+6-BA 2mg/L+2,4-D 2mg/L+ NAA 0.3mg/L and the callus induction rate was 81.28%.6-BA was also the main factor influencing callus proliferation,the effect sequence of four hormones was 6-BA＞NAA＞KT＞2,4-D,the optimal callus proliferation medium was MS+6-BA 1mg/L+KT 1mg/L+2,4-D 0.2mg/L+ NAA0.05mg/L and the callus proliferation rate was 52.71%.GA was a leading factor on the adventitious bud induction,the effect sequence of four hormones was GA＞6-BA＞NAA,the optimum medium was MS+6-BA 1mg/L+NAA 0.1mg/L+GA 0.5mg/L,and the adventitious bud induction rate was 78.21%;IAA was a major factor on the adventitious roots induction,the effect sequence of four hormones was IAA＞MS＞sucrose＞NAA,the optimum adventitious roots induction medium was 1/2MS+ NAA 0.3mg/L+sucrose 20g/L,and the adventitiousroots induction rate was 78.21%.
