Abstract: Objective: To identify the underlying mechanisms in the protective effects of Danzhixiaoyao Powder(DZXYP) on polycystic ovary syndrome rat model.Methods: PCOS was introduced by subcutaneous implantation of 18-methyl norethisterone silica gel stick combined with subcutaneous injection of human chorionic gonadotrophin.Two-dimensional polyacrylamide gel electrophoresis(2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS) were used to identify differentially expressed proteins.Immunohistochemistry,real time quantitative polymerase chain reaction(RQ-PCR) and western blot were performed to verify the results above.Results: DZXYP ameliorated ovary enlargement and follicle dilatation.Among eitht successfully identified proteins that were differentially expressed,β-arrestin 2(β-arr2) was upregulated in DZXYP pretreated PCOS group when compared with PCOS group.Conclusion: DXR could alleviate PCOS which might be related to increased expression of β-arr2.
