Abstract: Objective:To investigate the apoptosis-inducing effects of realgar nanoparticle（nano-realgar） on lung cancer A549 cells and the cancer stem cells（CSC）.Methods: Preparation of nano-realgar was mechanical milled using a high-energy planetary ball mill.MTT colormetric assay was used to detect the proliferating activity of A549 cells,Flow cytometry（FCM） was employed to assess the expression of intracellular P-glycoprotein（P-gp）,breast cancer resistance protein（BCRP） and the activity of Caspase-3,and the apoptosis of the A549 cells and their CSC（CD133+ cells） were assessed with Annexin V/PI double staining.Results: Nano-realgar significantly inhibited the proliferation of A549 cells and induced to apoptosis.Treated with 50 g/ml and 100 g/ml of nano-realgar for 48h,the apoptosis rate of the cells by Annexin V/PI staining was 12.59% and 69.19%,and the positive rates of activated Caspase-3 increased from（2.25±0.17）% to（3.84±0.63）% and（7.35±0.325） %,respectively.Administration the with 20 g/ml,50 g/ml and 100 g/ml nano-realgar for 48h,the relative proportion of CSC in overall A549 cell population obviously increased,meanwhile the apoptotic cells（CD133+ Annexin V+/PI+/-） in CSC population also gradually increased from（0.10±0.06）% to（4.28±0.42）%,（9.17±1.11）% and（30.71±2.82）%,respectively.The sensitivity of the CSC to nano-realgar was clearly lower than that of A549 cell population.The expression of BCRP and P-gp in A549 cells rarely changed after nano-realgar treatment.Conclusion: Realgar nanoparticle can effectively induce lung cancer A549 cells and their cancer stem cells to apoptosis.
