Microwave hyperthermia enhancing chemotherapeutic efficacy in retinoblastoma cells in vitroChinese Full Text

Abstract: Objective To study the ability of microwave hyperthermia in enhancing chemotherapeutic efficacy of doxorubicin( DOX) against human retinoblastoma Y79 cells in vitro. Methods MTT method was used to detect the viability of Y79 cells,cellular uptake of Y79 cells was determined by fluorescence microscopy,cell apoptosis was detected by flow cytometry,and spectrophotometric method was used to detect the expression of Caspase-3 in Y79 cells. The experiments were divided into 4 groups: blank control group,DOX group,microwave treatment group and DOX combined with microwave treatment group,and these groups were experimented,respectively. Results The Y79 cells exposed to DOX with microwave treatment showed a significant decrease in cell viability which compared with other groups( all P < 0. 05). The cellular uptake results showed that the cells increased permeability to DOX immediately with the addition of microwave compared with DOX alone,which continued to increase over 60 minutes. Apoptotic rate of cells treated with DOX and microwave hyperthermia was( 49. 08 ± 1. 02) %,and it could enhance the apoptosis rate compared to other experimental groups( all P < 0. 01). Meanwhile,the activity of Caspase-3 in cells of blank control group,DOX group,microwave treatment group and DOX combined with microwave treatment group were 2. 88 ± 0. 16,1. 61 ± 0.09,4. 67 ± 0. 27 and 1. 00 ± 0. 05,respectively,DOX with microwave treatment could enhance the activity of Caspase-3 in cells,there were statistical differences compared with other groups( all P < 0. 05). Conclusion Microwave hyperthermia can enhance the chemotherapeutic efficacy against retinoblastoma Y79 cells in vitro,DOX combined with microwave treatment is a promising approach for retinoblastoma.
