Abstract: Pdx1(pancreatic and duodenal homeobox 1) is one of the homeodomain transcription factors, and it is essential in pancreas development and islet β-cell function. It has been reported that homozygous deficiency of Pdx1 in mice leads to pancreatic agenesis. Pdx1 can also regulate β-cell maturation and differentiation by cooperating with other factors or cis-acting. In addition, Pdx1 increases insulin secretion through directly binding to and activating insulin promoter. In recent studies, Pdx1 is proved to play an important role in generation of β-like cells, gene editing and drug development. Therefore, this article mainly reviews the role and molecular mechanisms of Pdx1 in pancreas development and diabetes.
