The symbiotic and isolated culture characteristics of DSE in roots of Ammopiptanthus mongolicusChinese Full Text

Abstract: To understand the ecological significance of dark septate endophyte(DSE)associations in arid lands,the spatial dynamic of DSE fungi were surveyed in three plots including Yinchuan,Shapotou and Minqin located in northwestern sandland,China,in July 2013.Soil samples were collected in the rhizosphere of Ammopiptanthus mongolicus and were divided into five depth intervals,0-10,10-20,20-30,30-40 and 40-50 cm,respectively.The results showed that A.mongolicus could be highly infected by DSE and form a classical type of septate hyphae and microsclerotia,most of microsclerotia was discrete distribution and polymeric type was rare in Yinchuan.The proportion of polymeric type increased in Shapotou and Minqin.The ratio of microsclerotia/root length had significant differences between Yinchuan and Shapotou.The ratio of microsclerotia/root area had significant differences at the only10-20 cm depth among Shapotou and Yinchuan and Minqin.The highest hyphal infection was at 0-20 cm depth,and that in Minqin was significant lower than in Yinchuan and Shapotou.Microsclerotia infection was higher at 20-30 cm depth.The highest total infection and intensity were at 0-30 cm depth,there was a significant difference at the same depth among Minqin and Yinchuan and Shapotou.The 10 strains of DSE were isolated from roots,5strains could sporulate and the other strains could not sporulate,more often than not the asporulated strains were isolated from 0-10 cm depth and the sporulated strains were isolated from 20-30 cm and 40-50 cm depths.
